摘 要:目的探讨Mahoganoid(Md)基因在小鼠精子第一发生波中的动态表达以及其基因缺失对雄性小鼠生殖力的影响。方法采用荧光实时定量聚合酶链反应(Real-time PCR)相对定量法检测Mahoganoid mRNA在7、14、21、28、35、60d龄C3H/HeJ雄鼠睾丸中的表达;对Md基因缺失纯合子(Md^(nc))及C3H/HeJ雄鼠精子密度,精子活力,异常精子形态百分率分析,并通过交配实验观察其使正常雌鼠受孕分娩情况。结果 Mahoganoid mRNA在7d龄C3H/HeJ雄鼠睾丸有低表达,14 d表达开始上调,21 d上升趋势明显,35 d时表达水平为60 d 91%;Md^(nc)可以与雌鼠交配,但无幼鼠出生;Md^(nc)雄鼠精子快速前向运动百分比明显低于C3H/HeJ雄鼠精子(P<0.01)。结论 Mahoganoid基因与精子发生相关,Mahoganoid基因缺失会导致雄性小鼠精子活力下降,影响其生殖能力。Objective:To investigate the dynamic gene expression of Mahoganoid in mouse testis during spermatogenesis and its effect on fertility in male mice. Methods:The Mahoganoid mRNA levels in the mouse testes at postnatal days of 7, 14, 21, 28, 35 and 60 were measured by real-time reverse transcription-polymerase chain reaction. The sperm concentration, morphology and motility and mating assessment of both Mahoganoid-deficient (Md-/-) and wild-type (Md+/+) male mice were analyzed. Results. The expression of Mahoganoid mRNA in mouse testes was low on day 1, then increased gradually from day 7, with an obvious increase on day 21. The sperm actively progressively motility of Md-/ -mouse was obviously decreased when compared to that of Md+/+ mouse (P〈0.01), and the Md-/- male mice could mate with the normal females but no newborn mice were delivered. Conclusions:This study indicates that the Mahogunin deficiency in male mouse could result in the decrease of sperm actively progressively motility, which has an effect on the male fertility.
