摘 要:目的观察鼻敏片对实验性变应性鼻炎的疗效及可能的药理机制。方法50只Wistar大鼠,随机分为健康组、模型组、鼻敏片低剂量组、鼻敏片高剂量组。采用卵清蛋白制备变应性鼻炎模型,分别应用不同剂量鼻敏片及对照品进行处理,比较观察处理前后鼻部症状评分结果、鼻分泌物涂片和血涂片嗜酸性粒细胞计数结果,并取鼻黏膜进行免疫组化染色,分析比较各组鼻黏膜组织中P物质(Substancep,SP)和NGF(神经性因子,Nerve gracth factor,NGF)的表达活性差异。结果鼻敏片处理组大鼠鼻部症状减轻,P物质和NGF含量降低,症状表现与P物质和NGF含量变化呈正相关。结论P物质、NGF含量的异常变化是变应性鼻炎发病的重要因素。通过影响NGF、SP的含量水平,鼻敏片可以减轻变应性鼻炎症状。Objective To investigate the effects of Bimin Tablets (BMT) on the expressive activities of substance P (SP) and nerve growth factor (NGF) in nasal mucosa among modeling rats of allergic rhinitis and the correlation with therapeutic effect on the lesion. Methods Thirty Wistar rats were prepared as allergic rhinitis models at first by the procedures of ovalbumin, and then, they were randomly divided into 3 groups following the modeling procedures finished successfully, i.e. modeling controlling group (MCG), experimental group1 (EG1) treated with BMT at higher dosage and experimental group 2 (EG2) treated with BMT at lower dosage, with 10 rats in each. Another 10 healthy rats were taken as the healthy controlling group (HCG). Before and after treatment with various reagents, blood and nasal secretary samples were taken from all animals for eosinophils counting. By the end of experiment, al rats were put into death to take samples of nasal mucosa for immunohistochemistry to detect the expressive activities of SP and NGF, followed a comparative analysis on their differences in expressive activities among different groups. Results Among the animals treated by BMT, all showed significantly reduced allergic nasal symptoms and decreased expressive activities of SP and NGF in nasal mucosa, with a positive correlation of symptom manifesting magnitude with the expressive levels of SP and NGE Conclusions Abnormal changes in the expressive levels of SP and NGF in nasal mucosa may be one of the important pathogenesis factors, and BMT can give rise inhibitory effect on symptom manifestation of allergic rhinitis via intervening with the expressive levels of SP and NGE
