摘  要:目的:观察熊果酸(Ursolicacid,UA)保护糖尿病(DM)大鼠血管病变的作用,并探讨其对NF-κB蛋白表达的影响。方法:以STZ诱导DM大鼠模型,设DM模型组、UA两剂量治疗组(50mg/kg,25mg/kg)和氨基胍治疗组(100mg/kg),同时设正常对照组。给药8wk后,用血糖仪测定空腹血糖,试剂盒测定血清MDA和NO水平,HE染色观测主动脉的病理变化;同时,采用sABC法检测核转录因子(NF-kBp65)蛋白的表达。结果:UA50mg/kg治疗后DM大鼠血糖值降低,血清MDA和NO水平较模型组降低;病理切片和免疫组化结果显示UA治疗组血管损伤较模型减轻且NF-κB阳性表达明显低于模型组。结论:UA有保护DM大鼠血管损伤的作用,机制可能与UA抗氧化,从而抑制NF-kB的激活有关。AIM : The protection of ursolic acid for the aorta of diabetic rat and its effect on the expression of nuclear factor-KB were studied. Methods: Rats were intravenous injected with STZ at the dose of 60mg/kg. 3 days later, the rats whose blood glucose were over 11.1 mmol/L were regards as diabetic rats and were divided into 4 groups randomly: diabetic mellitus (DM) group, UA (50mg/kg and 25mg/kg) group, amgudine (100mg/kg) group. Normal group was arranged simultaneously. After been treated for 8 weeks, the levels of fasting blood glucose (FBC) , serum MDA and NO were measured. The pathohistology of aortic arch was examined and the positive expression of NF-κB in the aorta was tested by immunohistochemistry. Results: The levels of fast blood glucose( FBG), serum MDA and NO of UA group (50mg/kg) decreased and there were significant compared with DM group ( P 〈 0.01 ). HE staining demonstrated the injury of aorta was attenuated and the activation of NF-κB was inhibited in UA group. Conclusion: The protection of ursolic acid for the aorta of diabetic rat should be related to the inhibition to reactive oxide stress (ROS) and activation of NF-κB and attenuate diabetic vascular complications in DM rats.
