摘 要:目的:观察五灵脂(Faeces Trogopterorum,FT)水提取物对实验性动脉粥样硬化(artherosclerosis,AS)大鼠细胞间黏附分子-1(intercellular adhesion molecule-1,ICAM-1)表达及动脉内膜病变的影响,探讨FT抗AS炎症作用的机制。方法:采用高脂饮食结合腹腔注射维生素D3注射液复制动脉粥样硬化模型,模型复制成功后给予FT水提取物2.5g/kg、10g/kg灌胃干预。用药6周后采用免疫组化技术观察主动脉ICAM-1表达,反转录聚合酶链反应(reverse transcriptase-polymerase chain reaction,RT-PCR)检测主动脉ICAM-1 mRNA表达,透射电镜观察主动脉内膜病变情况。结果:同模型组比较,五灵脂水提取物能抑制大鼠主动脉ICAM-1及ICAM-1 mRNA表达,高剂量作用较低剂量作用明显。电镜显示FT组大鼠动脉病变轻于模型组。结论:FT能降低实验性动脉粥样硬化大鼠ICAM-1表达,减轻血管内皮病变程度,这可能是其抗AS炎症的分子机制之一。Objective :To explore the mechanism of Faces Trogopterus (FT) anti-atherogenesis inflammatory through observing the effects of water extracts of FT on the expression of intercellular adhesion molecule-1 in the experimental artherosclerosis rats. Methods : Sixty healthy male clean Wistar rats were randomly divided into 4 groups of 15 rats per group: model group,control group, FT I group( FT I ) ,FT Ⅱ group( Ⅱ ). The control group was fed standard feedstuff. The model, FT I and Fr Ⅱ group were duplicated by feeding high cholesterol diet and peritoneal injecting vitamin D3. Ten weeks later, FT I and FT Ⅱ group were interfered by intragastric administration with different dose(2.5 g/ kg, 10 g/kg) of the water extracts of FT for six weeks. At last, the expression of intercellular adhesion molecule-1 proteinum and mRNA in the endarterium of arterosclerotic rats were detected by immunohistochemical technique and reverse transcriptasc-polymerase chain reaction; the pathological changes in the endarterium of arterosclerotic rats was observed by transmission electron microscope. Results: The AS model was successfully duplicated. Compared with the model group, the expression of intercellular adhesion molecule-1 proteinum and mRNA in the end- arterium of the rats was significantly lower( P 〈 0.01 or P 〈 0.05 ). Transmission electron microscope result was that the endothelial damage of the aortas in FT I and FT Ⅱ groups was less severe than that in model group. Conclusions: FT can downregnalte the expression of ICAM-1 in the artery of experimental arterosclerotic rats, which may account for the anti-arteriosclerosis inflammatory effects of FT.
