摘  要:目的:观察毛葡萄叶水提物对FeCl3诱导大鼠血栓形成的影响及主要作用机制。方法:预防性给药10天后用FeCl3诱导大鼠颈总动脉血栓形成模型观察血栓形成,测定血栓湿重,HE染色观察血栓的形态学变化;放射免疫法测定血栓素(TXB2)、前列环素(6-keto-PGF1α)、内皮素(ET)变化,探讨其作用机理。结果:毛葡萄叶水提物对FeCl3诱导大鼠血栓湿重明显减轻;血浆TXB2、ET降低,6-keto-PGF1α水平升高。结论:毛葡萄叶水提物有明显抗血栓形成作用,其机制与血管ET及血液TXB2和6-keto-PGF1α含量水平有关。Objective: To investigate whether the Vitis quinquangularis Rehd extract(VRE) has a effect on anti-thrombosis induced by FeCl and its mechanism. Methods: After giving prophylactic drug for 10d, thrombi model were induced by FeCl3 on carotid artery of different experi- menatal rats. Morphology of thrombus was observed by HE staining and thrombotic weight was also detected. The content of TXB2, 6-keto- PGF1α and endothelin(ET) in plasma were determined by radioimmunoassay to study the mechanism. Results: It was shown that the Vitis quinquangularis Rehd extract had significantly inhibited thrombosis induced by FeCI3 and reducd the thrombotic weight. Meanwhile, it had down regulative the level of TXB2 and ET and up regulative the level of 6-keto-PGF1α in plasma. Conclusion : It is suggested that VRE has a effect on anti-thrombosis induced by FeCl3 , its mechanism may be associated with the content of TXB2 and 6-keto-PGF1α in plasma and ET from vessel, which had be provided an evidence of the exploiting a new anti-thromosis for clinical application .
