摘  要:目的:探讨碱性成纤维细胞生长因子(bFGF)联合骨髓间充质干细胞(MSCs)构建组织工程瓣膜(TEHV),能否在保证生物学性能不变的前提下,缩短MSCs体外构建TEHV的时间。方法:分离培养大鼠MSCs,进行细胞鉴定后种植于去细胞猪主动脉瓣膜支架上,再进行分组于体外构建TEHV。在培养基中加入5μg/L的bFGF分别培养7 d、14 d做为实验组A和实验组B,实验组C除培养液中不添加bFGF外,其余同实验组B。对照组为大鼠肌成纤维细胞构建的TEHV。各组TEHV进行形态学观察,DNA和羟脯氨酸含量检测。结果:MSCs表达CD29(94.82%)和CD44(93.59%),a-SMA和Vi mentin染色阳性,各组TEHV具有相似的形态学结构,DNA和羟脯氨酸含量的差异无统计学意义。结论:采用bFGF联合MSCs能在更短的时间内构建具有相同生物学性能的TEHV,从而缩短TEHV体外构建时间。Objective:To exploe the effect of the combined using bFGF and mesenchymal stem cells (MSCs)on the constructive time of the tissue engineered heart value (TEHV). Method.. MSCs of rats were isolated and cultured. After cell identification, MSCs were seeded onto decellularized aortic valve leaflet scaffold to constuct TEHV in vitro in each group. TEHV were cutlured with DMEM contained 5 ng/ml bFGF in vitro for 7 d in group A or 14 d in group B. In group C, TEHV were cutlured only with DMEM for 14 d. In control group, TEHV were constructed by using myofibroblasts as seed cells. TEHV in each group were examined morphologically and biochemically (DNA and 4 hydroxyproline). Result:MSCs expressed CD29 (94.82%) and CD44 (93.59%), and revealed a strong expression of a-SMA and vimentin. TEHV in each group showed simliar morphologcical structure. And the differences of DNA and hydroxyproline contents between each group have no statistical significance.Conclusion:Construction of TEHV by combined using bFGF and MSCs can shorten the time of construction in vitro with the same biological properties.
