摘  要:[目的]探讨hOGG1Ser326Cys多态与食管癌易感性、临床病理特性之间的关系。[方法]选取河南省新乡市188例食管癌患者和203例正常对照作为研究对象,采用病例-对照研究方法,应用多聚酶链反应-单链构象多态(polymerase chain reaction-single strand conformation polymorphism,PCR-SSCP)技术,分析两组人群hOGG1基因第326位点Ser/Ser、Ser/Cys和Cys/Cys基因型的分布,以及各基因型在不同临床病理特性中的分布差异。[结果]hOGG1三种基因型(Ser/Ser、Ser/Cys和Cys/Cys)在两组人群中的分布无统计学差异(χ2=2.13,P=0.344);在食管鳞状细胞癌组织学分型中,Ⅲ级与Ⅰ级相比,hOGG1Cys/Cys基因型在分化程度差的食管癌中的分布明显增加(OR=3.69,95%CI:1.16 ̄11.76)。[结论]hOGG1Cys/Cys基因型与食管鳞状细胞癌组织低分化相关,提示hOGG1活性降低可能预示食管癌预后不良。[ Objective ] To investigate if hOGG1Ser326Cys polymorphism is associated with esophageal cancer susceptibility, metastasis of esophageal carcinoma and histological differentiation. [ Methods ] The subjects comprised 188 cases and 203 healthy individuals as control from Xinxiang city, Henan Province. hOGG1 gene was genotyped by polymerase chain reaction-single strand conformation ( PCR-SSCP ). [ Results ] The distribution of hOGG1 Ser326Cys genotype among controls was not different from that among eases (X^2=2.13, P=0.344 ). hOGG1 Ser326Cys polymorphism was associated with the differentiation of esophageal carcinoma, The genotype of hOGG1 Cys/Cys^326 significantly increased the risk of poor differentiation of esophageal carcinoma( P=0.027, OR=3.69, 95%CI: 1.16-11.76 ). [ Conclusion ] This study showed that Cys/Cys genotype in hOGG1 gene played an important role in the differentiation of esophageal carcinoma, and the hOGG1 326Ser→Cys polymorphism might predict the prognosis of esophageal carcinoma.
