摘 要:[目的]观察长期应激结合高脂饲料对大鼠胸主动脉的损伤作用。[方法]雄性Wistar大鼠24只,随机分为4组,每组6只,分别为对照组(C组)、高脂饲料喂养组(HC组)、单纯足底电刺激并辅以噪声刺激组(CS组)及高脂饲料结合应激组(HC+CS组)。持续12周实验结束后取血,取胸主动脉。检测大鼠血清中总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)、游离脂肪酸(FFA)、超氧化物歧化酶(SOD)、丙二醛(MDA)、超氧阴离子(O2-.);采用放免法检测血清中肿瘤坏死因子(TNF-α)、ELISA法检测氧化型低密度脂蛋白(ox-LDL),电镜观察胸主动脉病理变化。[结果]血清学指标检测显示HC+CS组与HC组及CS组有明显的区别:出现了更明显的血脂紊乱(TC、FFA、LDL升高,HDL下降,尤其ox-LDL升高很明显),炎症反应(血清TNF-α升高),氧化应激(SOD活力明显下降,血清MDA和O2-.水平明显升高);同时电镜观察发现HC+CS组的胸主动脉也出现了明显损伤,而HC组和CS组的损伤程度明显较轻。[结论]长期应激结合高脂饲料有协同作用,可促进大鼠胸主动脉的损伤。[ Objective ] To investigate the effect of combination of high fat diet and stress on thoracic aorta injury in rats by long term stimulating. [ Methods ] Twenty-four male Wistar rats were divided into 4groups( 6each )randomly. The control group( C group )was fed with standard rat chow diet, the high fat dietary group( HC group )was fed with high fat diet, the chronic stress group( CS group )was fed with standard rat chow diet and stimulated by electric footshock and noise stress, the high fat dietary with chronic stress group( HC+CS group )was fed with high fat diet and stimulated by electric footshock and noise stress. After treated for 12weeks, serum biochemical indices including total cholesterol ( TC ), triglycerid ( TG ), high density lipoprotein ( HDL ), low density lipoprotein ( LDL ), free fatty acid( FFA ), superoxide dismutase( SOD ), malondialdehyde( MDA ), and O^-2 were measured.The level of TNF-α in serum was measured by radioimmunoassay kit and oxidative-loco density lipoprotein (α-LDL ) in serum was measured by the enzyme-linked immunosorbent assay( ELISA ). The rats were sacrificed and the thoracic aorta was collected for electron microscopic observation. [ Results ] The HC+CS group showed more evident dyslipidaemia( increased TC, FFA, LDL and especially α-LDL; decreased HDL ), inflammation( elevated TNF-α in serum )and oxidative stress( decreased SOD activity and increased MDA and O^-2 than HC group and CS group. Electron microscopic observation showed the HC+CS group had more severe thoracic aorta injury that other two groups. [ Conclusion ] The results suggested that the combination of high fat diet and chronic stress had synergistic effect in producing thoracic aorta injury.
