摘 要:目的:筛选人胎大脑皮质细胞冷冻保存的适宜条件。方法:将胎龄4~6月人胎大脑皮质制成细胞悬液并分别保存于4℃和液氮中,分别于保存第1d,5d,15d和30d用台盼蓝染色法观察细胞存活率。结果:①胎龄4~6M人胎大脑皮质细胞在冷存后均可存活,以胎龄4M细胞存活率较高。②冷存1d~5d时,4℃保存的细胞存活率高于液氮保存的细胞存活率,但随着保存时间的延长细胞存活率显著下降。③冷存1~30d时,液氮保存的细胞存活率无明显变化,且在15d和30d时细胞存活率明显高于4℃保存者。结论:人胎大脑皮质细胞的保存条件以胎龄4M、4℃短期保存和慢速冷冻、快速复温的液氮长期保存为宜。Objective: To screen out the optimal condition for cryopreservation of the cerebral cortical cells in the human fetus.Methods: The cerebral cortex of the human fetus from 4 to 6 months was made into the cell suspension and stored in 4℃ and liquid nitrogen respectively,then the survival rate of the cells was observed at 1d,5d,15d and 30d after cryopreserving respectively with trypan blue staining method. Results: ①The cells of the cerebral cortex in the human fetus from 4 to 6 months might survive after cryopreserving and the survival rate in the fetus aged 4 month was higher than that in the fetus aged 5 and 6 months. ②The survival rate of the cells stored in 4℃ was higher than that in liquid nitrogen at 1d and 5d after cryopreserving,but decreased remarkably with the lengthening of the storing time. ③The change of suevival rate of the cells stored in liquid nitrogen was not obvious from 1d to 30d,and the survival rate of the cells stored in liquid nitrogen was higher than that in 4℃ at 15d and 30d after cryopreserving.Conclusion: The optimal condition for cryopreservation of cerebral cortical cells was the fetal aged 4 month,stored in 4℃ for a short-term or stored in liquid nitrogen for a long-term.
