摘  要:目的:建立一种快速简便的的反相离子对高效液相色谱法测定血清中异烟肼(INH)浓度。方法:用SpherisorbSiO2(5μ,250mm×46mm)色谱柱,以甲醇:20mmol/LIPR-B8=15∶85(V∶V)为流动相,紫外检测器检测,波长254nm,烟酰胺(NA)为内标物。结果:在此色谱条件下,INH与NA的保留时间分别为7817min和6872min。在05~200μg/ml的浓度范围内,线性关系良好(r=09995)。方法日内差异219%,日间差异451%,回收率933%,最低检测浓度50ng/ml。结论:该法灵敏度高,特异性好,不需提取过程,大大减低了测定的工作量,对日常血药浓度监测和药代动力学工作的开展,具有极大的优越性。Objective:An ionpair HPLC method for the quantitative analysis of isoniazid in human serum was described.Methods:High performance liquid chromatograph Model LC10 A (Shimadzu ,Japan) with a 254 nm wavelength uvdetector and Spherisorb SiO2 column (5u,250×4.6 mm) was used.The mobile phase of methanol and 2.0 mmol/L IPRB8(15:85;V:V,pH 2.5) was pumped at 1.0 ml/min through the column.The temperature of column box was 40℃ and the detector was operated at 0.016 AuFs.Nicotinic amide was used as the internal standard .Results:The retention time for isoniazid and nicotinic amide was 7.817 min ,6.872 min,respectively.Standard curve was linear in the concentration range of 0.5 to 20.0 μg/ml for isoniazid .The recovery rate was 93.3%,withinday RSD was 2.19%,interday RSD was 4.51%,the minimum quantification concentration was 50 ng/ml.Conclusion:This method is rapid and convenient for monitoring the plasma concentration of isoniazid.
