鳖甲煎丸对人肾小球系膜细胞干预作用的研究

摘　　要：目的:探讨中药鳖甲煎丸药物血清对人肾小球系膜细胞(MCs)增殖和凋亡作用的影响。方法:应用细胞培养技术,进行人MCs培养,采取血清药理学方法,制备鳖甲煎丸药物血清,利用脂多糖(LPS)为刺激因子,将培养的MCs分为空白对照组(简称"空白组");鳖甲煎组;脂多糖组;脂多糖鳖甲煎组(简称"脂甲组")。应用四甲基偶氮唑蓝(MTT)法和流式细胞仪检测MCs增殖、凋亡和细胞周期。结果:LPS刺激后,增殖指数明显提高,与空白组比较有明显差异(P<0.01);鳖甲煎丸药物血清能明显抑制人MCs增殖(P<0.01),并且使多数MCs阻滞在G0/G1期,能诱导人MCs发生凋亡。结论:抑制MCs增殖和促进凋亡可能是鳖甲煎丸延缓慢性肾脏疾病进展的部分作用机制。Objective： To study the role of Biejia Jian Pill（BJJP） on human mesangial cells in culture. Method： We adopt cell culture technique and blood serum pharmacology method in experiment. Mesangial cells were stimulated by hpopolysaccharide（LPS）. There are four groups： control group; BJJP group; LPS group and LPS BJJP group. The MCs proliferation, apoptosis, and cell cycle of HMCs were measured by MTT assay and flow cytometry respectively. Result： After mesangial cells were stimulated by LPS,proliferation of mesangial cells were increased than that of control group （ P 〈 0.01 ）. BJJP ean significantly inhibit proliferation of mesangial cells compared with control group （ P 〈0.01 ） and delay the entry of cell cycle at G0/G1 period and induce apoptosis of HMCs. Conclusion ： Proliferation inhibition and apoptosis induction of HMCs are some of the mechanisms of BJJP in delaying glomerular sclerosis progression.
