摘  要:目的观察糖尿病大鼠肾组织P-选择素(CD62P)、细胞间黏附分子-1(ICAM-1)的表达及辛伐他汀的影响。初步探讨辛伐他汀对糖尿病大鼠肾组织的保护作用机制。方法将48只雄性Wistar大鼠随机分为正常对照组(NC)、糖尿病组(DM)和辛伐他汀治疗组(D),DM组和D组腹腔注射链尿佐菌素(STZ)建立糖尿病模型,成模后8周、12周免疫组化法测定CD62P、ICAM-1水平;用放射免疫分析法测大鼠24h尿白蛋白排泄率。结果与NC组相比,DM及D组CD62P、ICAM-1表达增加(P<0.01);D组CD62P、ICAM-1表达低于DM组(P<0.01);并且CD62P、ICAM-1水平与尿白蛋白排泄率呈正相关。结论辛伐他汀通过降低CD62P、ICAM-1的表达,减少血管内皮损伤,减轻肾脏炎症反应及微血栓形成,进而对肾脏起到保护作用。Objective To observe the expression of P-selectin (CD62P)and Intercellular adhesion molecule-1 (ICAM-1 )on diabetic rats, and investigate the mechanism of renal protective effect of Simvastatin on diabetic rat kidney. .Methods Forty-eight rats were assigned to normal control group(NC),diabetic rat group(DM) and Simvastatin treatment group(D). Immunohistochemistry were used to examine the expression of CD62P and ICAM-1 in 8 and 12 weeks in kidney of rats. Observe the expression of urinary albumin excretion rate in 24 hours of rats. Results In comparison with normal group ,the expression of P-selectin and ICAM-1 in diabetic rat and simvastatin treatment groups was markedly increased (P〈0.01). In comparison with diabetic rat group, simvastatin could significantly decrease the expression of P-selectin and ICAM-1 (P〈0.01). The relationship of P-selectin and ICAM-1 was positively correlated with urinary albumin excretion rate.Conclusion Simvastatin may play a protective role of the kidney by down-regulating P-selectin and ICAM-land alleviating renal injury in diabetic rat.
