摘  要:目的探讨异氟醚延迟预处理对心肌缺血再灌注损伤兔心肌蛋白质组的影响。方法成年新西兰大白兔8只,雌雄不拘,体重2.0—2.5kg,随机分为2组(n=4):缺血再灌注组(IR组)和异氟醚延迟预处理组(IDP组)。采用结扎左冠状动脉前降支40min,再灌注120min的方法建立缺血再灌注模型。IDP组持续吸入2%异氟醚2h,24h后进行缺血再灌注。于再灌注结束即刻,取左心室前壁心肌组织,进行双向凝胶电泳和质谱分析。结果与IR组比较,IDP组差异表达的蛋白质有13个,其中10个蛋白质表达上调,3个蛋白质表达下调(P〈0.05)。质谱分析得到13张肽质量指纹图谱,并成功鉴定出了其中11个蛋白质。结论异氟醚延迟预处理可减轻兔心肌缺血再灌注损伤,可能与心肌蛋白质组的变化有关。Objective To investigate the effects of isoflurane delayed preconditioning (IDP) on myocardial proteome in rabbits with myocardial ischemia-reperfusion (I/R) injury. Methods Eight New Zealand white rabbits of both sexes weighing 2.0-2.5 kg were randomly divided into 2 groups (n = 4 each) : I/R group and IDP group. Myocardial I/R was induced by occlusion of left anterior descending artery for 40 min followed by 120 min reperfusion. In group IDP, the animals inhaled 2% isoflurane for 2 h, undergoing I/R 24 h later. At the end of 120 min reperfusion, the myocardium of left ventricle anterior wall was removed for two-dimensional gel electrophoresis. The different protein spots were analyzed by means of mass chromatography. Results There were 13 different protein spots between group I/R and IDP. Of the 13 proteins, the expression of 10 spots was up-regulated and 3 spots down-regulated in quantity. Eleven protein spots of all spots were identified by means of MALDI-TOF-MS. Conclusion IDP can attenuate myocardial I/R injury in rabbits and it may be related to the alteration in proteome of myocardium.
