摘  要:目的探讨高糖对小鼠肾小球足细胞podocalyxin mRNA表达的影响。方法以体外培养的小鼠足细胞为研究对象,将细胞分组,即正常对照组(NG组):D-葡萄糖5.56mmol/L;高糖培养组(HG组);D-葡萄糖25.0mmol/L。应用逆转录-PCR技术,检测高糖培养不同时间点对足细胞表达podocalyxin mRNA的影响;Western blot印迹分析检测细胞转录因子WT-1蛋白变化。结果①逆转录-PCR技术显示随着高糖培养时间增加,HG组肾小球足细胞与WG组比较、以及HG组(第6、12、24h)组间比较,podocalyxin mRNA表达均减少(P〈0.05)。②培养的足细胞表达基础水平的WT-1蛋白,但在经高糖培养2h后其表达即开始下降(P〈0.05)。结论高糖培养使小鼠肾小球足细胞podocalyxin mRNA和转录因子WT-1蛋白表达下降。Objective To explore the regulatory effect of high concentration glucose on the expression of podocalyxin mRNA in cultured mouse podocytes. Methods Podocytes were cultured in vitro as study samples. The expression of pdocalyxin mRNA in mouse podocytes cultured in high concentration glucose was detected at different time points by reverse transcription-PCR (RT-PCR), and the changes of WT-1 protein were analyzed by Western blot. Results ①RT-PCR revealed that the expression of podocalyxin mRNA in mouse podocytes cultured in high concentration glucose was significantly reduced as compared with control group at different time points (P〈0. 05). With the prolongation of culture of the mouse podocytes under high concentration glucose, the expression of podocalyxin mRNA was decreased (P〈0. 05). ②The WT-1 protein baseline level in podocytes cultured in vitro was decreased after culture with high concentration glucose for 2 h (P〈0. 05). Conclusion Both the expression of podocalyxin mRNA and WT-1 protein in cultured mouse podocytes was declined after culture with high concentration glucose.
