金欣口服液对RSV感染人胚肺成纤维细胞β-干扰素mRNA和蛋白表达的影响  被引量:2

摘  要:目的探讨金欣口服液含药血清对呼吸道合胞病毒(RSV)感染人胚肺成纤维(HELF)细胞是否通过诱生β-干扰素影响感染细胞增殖。方法体外建立RSVA型LONG株攻击HELF细胞模型,设细胞对照组、病毒对照组、空白血清组、利巴韦林组和金欣口服液组。采用Real-time PCR及ELISA法检测各组细胞β-干扰素mRNA及蛋白的表达。结果与正常细胞(2-△△Ct值为1.79)比较,RSV感染24h后HELF细胞β-干扰素mRNA的表达量没有明显下降(2-△△Ct值为1),空白血清组、利巴韦林组、金欣口服液组分别为0.967,0.86,0.57,差异均无统计学意义(P>0.05)。采用先加药后感染病毒、先感染病毒后加药、单用含药血清干预3种加药方式,各组β-干扰素表达均为0ng/L。结论(1)RSV不刺激β-干扰素mRNA及蛋白的生成;(2)金欣口服液也不诱导β-干扰素mRNA及蛋白的生成,而金欣口服液含药血清可抑制感染细胞的增殖作用,可能存在其他的非依赖于β-干扰素的途径。Objective To study the effect of Jinxin oral liquid on expressions of mRNA and protein of interferon-β(IFN-β) in RSV-infeeted human embryonic lung fibroblast cells in vitro and to discuss the treatment mechanism of Jinxin oral liquid to the RSV infected cell proliferation. Methods We successfully established the cell model of RSV by the long strain(A type of RSV). We divided them into five groups,which were normal cell group,RSV group, blank serum group, ribavirin group and Jinxin oral liquid group. In the experiments, the real time PCR and ELISA were used to examine expressions of mRNA and protein of IFN-β in RSV-infeeted human embryonic lung fibroblast cells in different groups. Results The mRNA of IFN-β expression in human embryonic lung fibroblast cells did not significantly descend after being infected by RSV(the value of 2^-△△Ct equaled 1) compared with those in normal fibroblast cells(the value of 2^-△△Ct equaled 1.79)(P〉0.05). The relative value of blank serum group and ribavirin group was 0. 967 and 0. 86 (P〉0. 05). In the relative value of Jinxin oral liquid group,it was 0.57 (P〉0.05). Three prescription methods were adopted. First method was that the cells were added the medicine serum and then they were infected by RSV. The cells were infected by RSV and then were added the medicine serum in the second method. In the last method cells were interfered with the medicine serum alone. Unfortunately,the protein expression of IFN-9 in each group was 0 ng/L. Conclusion (1)RSV does not stimulate human embryonic lung fibroblast cells to express the mRNA and protein of IFN-β. (2) Jinxin oral liquid does not induce the mRNA and protein of IFN-β production. Jinxin oral liquid might inhibit the RSV infected cells proliferation. However, there might be other ways independent of IFN-β.
