高同型半胱氨酸血症对大鼠血脑屏障的影响

摘  要:目的:探讨高同型半胱氨酸血症(HHcy)对大鼠血脑屏障损伤的作用机制。方法:通过高蛋氨酸饮食制备HHcy大鼠模型(HHcy组),并设普通饲料饲养大鼠做对照。实验初始及24周末检测2组大鼠血浆总同型半胱氨酸(tHcy)及血清一氧化氮(NO),第24周末检测2组大鼠脑组织伊文思蓝含量及脑组织含水量,利用透射电镜观察血脑屏障超微结构。结果:HHcy组与对照组实验初始及第24周末血浆tHcy分别为(13.19±3.10)μmol/L、(13.37±3.18)μmol/L(P>0.05)及(22.51±4.47)μmol/L、(13.92±4.16)μmol/L(P<0.05)。2组实验初始及第24周末血清NO分别为(28.44±8.86)μmol/L、(27.11±7.81)μmol/L(P>0.05)及(12.74±5.73)μmol/L、(27.13±5.85)μmol/L(P<0.05)。第24周末HHcy组与对照组脑组织伊文思蓝含量及脑组织含水量分别为(7.37±1.07)μg/g、(5.90±0.46)μg/g及(78.10±0.31)%、(74.93±0.28)%(均P<0.05)。电镜示对照组血脑屏障结构正常,HHcy组脑血管内皮细胞结构欠清晰,基膜局部不规则增厚等变化。结论:HHcy可造成大鼠血脑屏障损伤,推测其机制与一氧化氮活性降低致血管内皮损伤有关。Objective: To investigate the mechanism of hyperhomocysteinemia (HHcy) on the blood-brain barrier permeability in rats. Methods: The patho-models of rats with hyperhomocysteinemia were constructed by feeding homomethionin diets, and rats in control group were fed with normal diets. Plasma total homocysteine (they) and serum nitric oxide were detected at the beginning and 24th week of the experiment. Evans blue and water content of brain were detected respectively, and ultramicrostructure of blood-brain barrier were observed with electron microscope at the 24th week of the experiment. Results: The level of plasma tHcy in HHcy group and control group were (13.19±3.10)umol/L vs (13.37±3.18)umol/L (P 〉 0.05) at the beginning of the experiment, while they were (22.51±4.47)umol/L vs (13.92±4.16)umol/L at the 24th week of the experiment (P 〈 0.05). The level of serum nitric oxide in both HHcy group and control group were (28.44±8.86)umol/L vs (27.11±7.81)umol/L at the beginning of the experiment (P 〉 0.05), while they were (12.74±5.73)umol/L vs (27.13±5.85)umol/L at the 24th week of the experiment (P 〈 0.05). Evans blue and water content of brain in HHey group and control group were (7.37±1.07)ug/g vs (5.90±0.46)ug/g and 78.10%±0.31% vs 74.93%±0.28% at the 24th week of the experiment (all P 〈 0.05). Electron microscopic features blood-brain barrier with normal appearance in control group, while the structure of capillary endothelial cells was irregular, and the basement membrane of cerebral capillary thicken locally were obvious. Conclusion: HHey may increase the permeability of blood-brain barrier in rats. The mechanism maybe correlate with endothelial dysfunction with decreased nitric oxide bioavailability.
