摘  要:目的:研究大鼠远位触液神经元(distal cerebrospinal fluid contacting neuron,dCSF-CN)是否参与颌面部炎性痛的痛觉调制过程。方法:利用颌面部福尔马林致痛模型,采用免疫组织化学双标及免疫电镜技术结合福尔马林行为学实验观察P物质(Substance P,SP)在dCSF-CN和三叉神经脊束核(spinal trigeminal nucleus,STN)的表达。SPF级雄性SD大鼠(体重290~310 g)12只随机分成两组(n=6)。麻醉后用立体定位侧脑室注射30%CB-HRP3μl逆行示踪标记dCSF-CN。48h后行颌面部福尔马林试验并观察行为学差异,福尔马林注射2h后灌注固定取材行组织学分析。结果:(1)dCSF-CN是脑干内的一类特别的神经元,其胞体位于室管膜内的脑实质、突起伸入脑室腔的脑脊液;(2)颌面部注射福尔马林诱导的急性炎性痛显著上调SP在dCSF-CN和STN的表达,并与其行为学表现相一致。结论:推测SP在dCSF-CN和STN的痛觉调制中扮演重要角色。Objective: To explore whether the distal cerebrospinal fluid contacting neuron (dCSF-CN) modulated the orofacial inflammatory pain in rats. Method: Twelve male Sprague-Dawley rats (290 -310 g, Grade SPF) were randomly divided into 2 groups. Rats were anesthetized and fixed in the stereotaxic instrument. A 3-μL volume of 30% CB-HRP to trace dCSF-CN was injected into one of the rats'lateral ventricles. Forty eight hours later nociception of the animals was assessed behaviorally with the rat orofacial formalin test. Two hours after formalin administration rats were sacrificed with deeply anesthesia and transcardially perfused. Brainstem segments of dCSF-CN and STN were removed by SP/CB-HRP dual-labeling with immunohistochemical procedures and colloidal gold immunoelectronmieroscopy. Results : ( 1 ) The dCSF-CN is a peculiar type of neuron whose body is in the parenchyma of the brain and processes extend into cerebrospinal fluid (CSF) in the cavity of the ventricle in the central nervous system; (2) Both SP-containing neurons in dCSF-CN and in STN are up-regulated significantly 2h after formalin injection. Conclusion: SP-expression in dCSF-CN and STN may be considered to play an important role in nociceptive processing.
