摘 要:目的探讨瘦素、纤连蛋白(FN)、基质金属蛋白酶-9(MMP-9)对人绒毛细胞滋养细胞侵袭性的影响。方法取孕6-10周人工流产妊娠绒毛,分离、培养细胞滋养细胞(CTB)并经光镜、免疫细胞化学鉴定;应用体外培养侵袭模型检测瘦素、FN、MMP-9及瘦素+FN对CTB侵袭能力的影响。结果瘦素、FN、MMP-9及瘦素+FN作用后,浸润穿过基底膜的CTB数均显著多于对照组(P均〈0.01),尤其是瘦素+FN。结论瘦素、FN、MMP-9能增强CTB的侵袭能力;瘦素与FN有协同作用。Objective To investigate the effects of leptin, fibronectin (FN) and MMP-9 and on the invasion of human cytotrophoblast(CTB). Methods CTB were isolated from chorionic villi at 6 - 10weeks pregnancy,valuated by light microscope and immunocytochemistry. The effects of leptin ,fibronectin and MMP-9 on the invasion of CTB were detected by using invasion model. Results The number of penetrated-membrane-cells in leptin, FN, MMP-9,1eptin + FN groups were signifi- cantly higher than that in the control group( P 〈 0.01 ). In particular which in the leptin + FN group. Conclusions Leptin, FN, MMP-9 can increase the invasion ability of CTB. Leptin and FN have synergy.
