摘 要:目的:研究丹参注射液对大鼠离体心脏能量代谢的调节作用。方法:采用大鼠离体心脏灌流技术,测定在添加或不添加脂肪酸的情况下,大鼠心脏中乙酰辅酶A(AcCoA)、辅酶A(CoA)、高能磷酸化合物、乳酸的含量,并观察给予丹参注射液和阳性药肝乐(DCA)干预后各指标的变化。结果:在不含脂肪酸的情况下,DCA和丹参注射液均显著提高大鼠心肌组织中AcCoA/CoA值;在含有脂肪酸的情况下,DCA对AcCoA/CoA值没有显著影响,而丹参注射液显著降低了AcCoA/CoA值;在含有脂肪酸的情况下,加入丹参注射液会导致DCA组大鼠心肌组织中AcCoA/CoA值降低。丹参注射液可使离体大鼠心脏中ATP含量升高,能荷值升高,乳酸含量降低。结论:丹参注射液抑制离体大鼠心脏中脂肪酸的氧化,提高心肌能量代谢的效率,起到心肌保护作用。Aim: To observe the regulation on rat cardiac energy metabolism by Danshen injection. Methods: Isolated hearts were perfused with or without fatty acid for 30 rain. At the end of the perfusion the hearts were pre- served in liquid N2 until being analyzed for CoA, acetyl CoA, AMP, ADP, ATP, and lactate. Changes to the various biomarkers were observed when Danshen injection and DCA were added. Results: Both DCA and Danshen injec- tion increased the ratio of AcCoA to CoA in the absence of fatty acid in the rat heart tissues. The addition of DCA lead to no significant alteration in the ratio of AcCoA to CoA in the presence of 0. 8 mmol/L palmitic acid. However, the addition of Danshen injection caused dramatic reduction in the ratio of AcCoA to CoA in the tissues. Significant increase in ATP, ADP and AEC levels in rat heart tissues were observed when Danshen injec- tion was added. The presence of fatty acid caused an increase in lactate level in rat hearts, which was reversed by the addition of Danshen injection. Conclusion: Danshen injection may indirectly increase cardiac glucose oxida- tion by inhibiting cardiac fatty acid oxidation, increase heart work efficiency and protect the heart.
