摘 要:目的:研究大黄酚对麻醉大鼠海马齿状回突触传递活动的影响。方法:采用在体细胞外记录长时程增强(LTP)的电生理学方法,观察侧脑室注射大黄酚(20,10,5ng/μL)对海马齿状回基础突触传递活动和高频刺激诱导LTP的影响,以及给予东莨菪碱后,大黄酚对大鼠海马齿状回高频刺激诱导LTP的影响。结果:大黄酚可剂量依赖性增强大鼠海马齿状回基础突触传递活动;对于高频刺激诱导LTP现象有增强作用;对抗东莨菪碱对高频刺激诱导LTP的抑制作用。结论:脑内静脉注射大黄酚可剂量依赖性增强海马齿状回基础突触传递活动和高频刺激所诱导的LTP,其作用机制可能与大黄酚改善胆碱能系统的功能有关。Aim: To observe the effect of chrysophanol on the synaptic transmission in the dentate gyrus of hippo- campus in rats. Methods: Using electrophysiological method of in vivo extracellular recording, the effects of in- tracerebroventricularly injection of chrysophanol(20, 10, 5 ng/μL) on the basal synaptie transmission activities, long-term potentiation (LTP) induced by high-frequency stimulation (HFS), and HFS-induced LTP after admin- istration of scopolamine in dentate gyms of hippocampus were studied. Results: In anesthetized rats, chrysophanol dose-dependently enhanced both the basic synaptie transmission and the magnitude of HFS-induced LTP in the dentate gyrus of hippocampus. Moreover, it rivaled the inhibitory action of cholinergic receptor-blocking drug sco- polamine on HFS-induced LTP. Conclusion: Chrysophanol dose-dependently enhanced the basic synaptic trans- mission and the magnitude of HFS-induced LTP in the dentate gyrus of hippoeampus in anesthetized rats. The mechanism may be related to the improve ment in the cholinergic system function by chrysophanol.
