摘 要:目的:研究非促分裂型酸性成纤维细胞生长因子(nmhaFGF)对缺血再灌注损伤心肌的保护作用。方法:大鼠冠脉左前降支闭塞30min后再灌注3h,再灌注前10min尾静脉注射不同剂量nmhaFGF。测定各组大鼠室性心动过速(VT)和心室颤动(VF)的潜伏期、持续时间和发生率,评定心律失常分数;再灌注24h后,测定血清AST、LDH、CK-MB含量,并计算大鼠心肌梗死面积。结果:静脉注射nmhaFGF(20,40,80μg/kg)能明显抑制缺血再灌注引起的心肌损伤。和模型组相比,nmhaFGF各组大鼠的VT和VF发生率减少,出现心律失常的潜伏期延迟,持续时间缩短;另外心肌梗死区占左室面积的百分比及梗死区占危险区百分比均有所降低;此外,血清心肌酶含量AST、LDH、CK-MB显著降低;以80μg/kgnmhaFGF作用较明显,其作用强度与8mg/kg普萘洛尔相当。结论:nmhaFGF对大鼠缺血再灌损伤心肌具有保护作用。Aim: To investigate the protective effect of non-mitogenetic human acidic fibroblast growth factor (nmhaFGF) against myocardial isehemia-reperfusion injury in rats. Methods: Myocardial ischemia-reperfusion injury was induced by 30-minute occlusion of left anterior descending coronary artery (LADCA) followed by 3-hr reperfusion. Viable doses of nmhaFGF were infused intravenously at 20 minutes after LADCA occlusion. The inci- dence and latency phase and duration of ventricular tachycardia (VT) and ventrieular fibrillation (VF) were mo- nitored, and the fraction of arrhythmia was reckoned; myocardial infarct area, the serum aspartate aminotransferase (AST) and lactate dehydrogenase (LDH) and MB isoenzyme of creatine kinase (CK-MB) content were detected at 24 hours after reperfusion. Results: Results showed that nmhaFGF (20, 40, 80 I^g/kg) could relieve myocardi- al injury induced by ischemia-reperfusion. Compared to the model group, mmhaFGF of the selected doses was re- sponsible for the decreased incidences in VT and VF, the delayed latency, the shortened duration and the reduced fraction of arrhythmia. In addition, the percentages of the infarct area relative to the total area of the left ventricle and the percentage of the infarct area relative to area at risk markedly decreased. Moreover, there were dramatic- ally decreases in AST and LDH and CK-MB contents in nmhaFGF groups there was no statistical difference be- tween nmhaFGF 80 μg/kg group and propranolol 8 mg/kg group. Conclusion: nmhaFGF could protect the myocardium against the damages induced by ischemia-reperfusion in rats.
