摘 要:目的考察水蛭素气雾给药对大鼠凝血功能及体内血栓形成的影响。方法测定大鼠凝血酶原时间(PT)、凝血酶时间(TT)、活化的部分凝血酶时间(aPTT),血浆纤维蛋白原(FIB)含量,观察水蛭素气雾给药的抗凝血作用;建立下腔静脉结扎模型和动-静脉旁路结扎模型,测定大鼠静脉及动脉血栓的湿重和干重。结果与正常对照组相比,水蛭素气雾给药中剂量和高剂量组(500 U.kg-1,1 000 U.kg-1)对PT,TT,aPTT有显著延长作用(P<0.05或P<0.01);对大鼠静脉血栓、动脉血小板血栓形成均有显著的抑制作用(P<0.05或P<0.01),对FIB无明显影响(P>0.05)。结论水蛭素气雾给药能够发挥其抗凝血作用。Objective To investigate the effects of hirudin by aerosol on blood coagulation function and thrombosis formation in rats. Methods To investigate the effects of hirudin aerosol preparation on prothrombin time(PT) , thrombin time(TT), activated partial thromboplastin time( aPTT), and the content of fibrinogen(FIB) in rats. Built up venous thrombosis model and arteriovenous shunt model to analysis the effect of hirudin by aerosol on venous thrombosis in rats. Results High and middle dosages of hirudin by aerosol could significantly prolong aPTT,TT,PT and decrease the weight of both venous and thrombocytic thrombus in rats( compared with NS, P 〈0.05 or P 〈0.01 ). However, they had no effect on the content of FIB in rats. Conclusion Hirudin aerosol preparation can reach an effective blood levels, has obvious effect on blood coagulation function and thrombosis formation in rats.
