摘  要:目的开发一种新技术从莲子心中分离纯化莲心总碱。方法对7种不同极性大孔吸附树脂静态吸附进行考察;采用正交实验通过对树脂动态吸附和洗脱进行研究。结果DA201树脂对莲心总碱的静态吸附性能最佳;得到了分离纯化莲心总生物碱的最优工艺条件为:在吸附实验中,上样液浓度为1.87 mg.ml-1,pH值为10,体积为8 BV,流速为2 ml.min-1。在洗脱实验中,乙醇洗脱剂的浓度为70%,pH值为3,体积为8 BV,流速为2 ml.min-1。结论DA201大孔吸附树脂对莲心总碱的分离纯化效果良好,树脂使用寿命长,具有较好的工业化前景。Objective To set up a new technique for purification of total alkaloids from embry of Nelumbo nucifera gaertn seed by macroporous resin. Methods Seven kinds of different macroporous resins were adopted to absorb total alkaloids from Nelumbo nucifera gaertn seed. The orthogonal test was used to analyze dynamic absorption and desorption. Results DA201 resin had the best absorption quantity. The optimum conditions were as follows absorption conditions- concentration 1.87 mg.ml^-1, pH 10, volume 8BV, velocity of flow 2 mg.ml^-1 ; desorption conditions : ethanol concentration 70% , pH3, volume 8BV, velocity of flow 2 mg .min^- 1. Conclusion It has good effect to separate total alkaloids from embryo of Nelumbo nucifera gaertn seed by macroporous resin DA201 which can be used for a long time. There is preferable industrialized foreground.
