摘  要:目的建立高效液相色谱法(HPLC)测定大鼠附睾组织中肉毒碱的含量。了解奥硝唑引起弱精子症模型动物附睾中影响精子成熟的标志性物质——肉毒碱含量的动态变化,探讨引发动物弱精子症的可能原因。方法用奥硝唑构建弱精子症模型,给药组分别给予200 mg/kg奥硝唑灌胃,空白对照组给予1%羧甲基纤维素钠(CMC-Na)灌胃。末次给药24 h后的不同时间用25%的乌拉坦溶液麻醉后取附睾。附睾组织匀浆用蛋白沉淀法进行样品处理,用HPLC法检测在用奥硝唑连续造模20 d期间大鼠附睾中肉毒碱的动态变化。结果奥硝唑可以使附睾中肉毒碱的含量在造模第2天迅速降低,在造模第5天达最低,并最终维持在4 300μg/ml水平,此水平与空白对照组相比约降低了24%。结论奥硝唑致动物弱精子症的原因可能是其细胞毒性影响了附睾的吸收功能,干扰附睾对肉毒碱的主动摄取,破坏精子成熟的环境,从而降低了精子的质量。Objective To develop a method for determining carnitine in rat's epididymis using HPLC. Methods Amblyspermatozoa animal model was established by ornidazole. Ornidazole(200 mg/kg) was given to the medication group by lavage, and 1% CMC-Na to the blank group by lavage. The rats were anesthetized with urethane 24 h after final lavage,and then epididymis was taken out. Epididymis homogenate was precipitated by acetonitrile and water. HPLC was used to determine the dynamic changes of carnitine in epididymis of rats during a period of 20 days. Results Ornidazole could quickly reduce the content of carnitine in epididymis of rats at 2nd day after creation of the model, reached the lowest at 5th day,and finally maintained at 4 300 μg/ml that was decreased by about 24% as compared with blank group. Conclusion Ornidazole affected the sperm quality first by interfering with the initiative uptake of carnitine by epididymis, then by changing the environment of mature sperm,at last resulting in the decline of sperm quality.
