摘 要:目的制备并检测异种骨支架材料和种子细胞的生物相容性、验证脂肪源间充质干细胞作为种子细胞及异种骨作为支架材料的可行性。方法制备异种骨支架材料,并与脂肪源间充质干细胞复合培养,分别做扫描电镜观察、细胞活性检测和碱性磷酸酶的测定,各组均以人同种异体骨支架材料作对照。结果复合培养4和10d后电镜下两种材料均见到脂肪源性间充质干细胞的存活并伸出伪足黏附在材料上;MTT法显示两组材料上的细胞增殖数均随时间的延长而呈增长趋势;与同种异体骨支架材料相比,异种支架材料内细胞碱性磷酸酶的变化趋势与其基本一致,均随时间的延长而增高。结论脂肪源间充质干细胞与异种骨支架材料有较好的生物相容性,两者复合构建组织化工程骨的前景值得进一步研究。Objective To study the compatibility between human adipose-derived mensenchymal stem cells and porcine bone scaffolds. Methods Porcine bone tissues were co-cultured with adipose-derived mesenchymal stem cells, and the complex was observed under scanning electron microscope. The viability and alkaline phosphatase (ALP) activity of the cells were examined with the cells co-cultured with human bone scaffold as the control. Results At 4 and 10 days after the co-culture, the adipose-derived mesenchymal stem cells were observed to extend pseudopodia to adhere to the two scaffold materials. MTT assay showed that the cell proliferation on both of the materials increased with time, and the two cell complexes exhibited similar pattern of changes in ALP activity. Conclusion As the seed cells, human adipose-derived mesenchymal stem cells exhibit good comparability with porcine bone scaffold, suggesting their potential of constructing tissue-engineered bone graft.
