摘 要:目的:观察粉防己碱(tetrandrine,Tet)对雄黄中毒大鼠的排砷作用。方法:取大鼠60只,随机分为正常对照组、雄黄中毒模型组(模型组)、雄黄+二巯基丙醇磺酸钠(DMPS)组、雄黄+Tet高、中、低剂量(Tet高、中、低剂量)组,每组10只。每组每日灌胃给予雄黄,各给药组分别给予不同剂量的药物腹腔注射,连续6周。给药期间收集大鼠24h尿液,以热消化-二乙氨基二硫代甲酸银(DDC-Ag)比色法的改良法测定尿砷含量,每周一次。停药后心脏取血以同样方法测定血砷含量。结果:各给药组大鼠24h尿砷含量与模型组比较,Tet高、中剂量组以及低剂量组前4周的尿砷差异有统计学意义(P<0.01),低剂量组的第5、6周尿砷量差异也有统计学意义(P<0.05);与DMPS组比较,仅低剂量组显示差异(P<0.05)。各给药组大鼠血砷含量与模型组比较,具有统计学意义(P<0.01);与DMPS组比较,仅低剂量组有统计学差异(P<0.05)。结论:Tet对雄黄中毒大鼠具有明显的促进砷排泄作用,其作用与剂量呈正比例关系。Objective:To investigate effects of tetrandrine(Tet) on eccritic function of arsenic in rats with realgar intoxication.Methods:Sixty rats were randomly divided into normal control group,model group with arsenic poisoning(model group),arsenic poisoning by sodium dimercaptopropane sulfonate(DMPS) treated group(DMPS),arsenic poisoning by Tet treated high dosage group(Tet high dose group),arsenic poisoning by Tet treated middle dose group(Tet middle dose group) and arsenic poisoning by Tet treated low dose group(Tet low dose group) in each group 10 rats.The realgar was taken by gavage(ig) for each group once a day for 6 weeks except the normal control group.At the same time,drugs were taken by intraperitoneal injection(ip) for 6 weeks.During the drugs administrated,the each group rats' 24h urine was collected to determine urine arsenic content in the improved DDC-Ag way once a week.Blood arsenic content which was taken from the rats' heart was determined in the same way after stopping administration.Results:The 24h urine arsenic content in the Tet high dose group,middle dose groups and low dose group had a significant difference compared with the model group(all P<0.01),with no statistical significance compared with the DMPS group except the Tet low dose group(P<0.05).The blood arsenic content in the 3 Tet dose groups was different compared with the model group(all P<0.01),and was significantly different only in the Tet low dose group compared with the DMPS group(P<0.05).Conclusion:It is demonstrated that Tet promotes arsenic excretion in rats with realgar poisoning and that Tet is in proportion to its dose.
