摘 要:目的观察过滤对血小板的影响,遴选适于过滤的血小板制品。方法对机采血小板、手工浓缩血小板和相应的两种冰冻血小板进行过滤,观察血小板回收率、白细胞去除率和血小板聚集率等多项血小板功能指标。结果机采血小板回收率为86.81%,手工浓缩血小板回收率仅为73.08%;两种冰冻血小板制品损失量达60%~70%;过滤后机采血小板达到去白细胞标准,而手工血小板未达到去白细胞标准;过滤对液态保存血小板制品的功能无明显影响,但手工浓缩血小板功能低于机采血小板。结论冰冻保存的血小板制品不适于去白细胞过滤;手工浓缩血小板制备工艺尚需进一步改进和提高,并研制与之相适应的去白细胞滤器;机采血小板质量较好,国产滤器可达到去白细胞标准,是当前适于滤除白细胞的血小板制品。Objective The effect of filtration on the platelet preparations was observed,and platelet preparations suited to platelet filtration were selected. Method The filtration effect of apheresis platelet, platelets concentration and their relative frozen platelets was studied while observing the platelet recovery rate,leukocyte removal rate, platelet concentration rate and various platelet function indexes. Results The platelet recovery rate for apheresis platelet was 86.81%, while manual concentrated platelet was only 73.08%. The platelet loss rate for two frozen platelet preparations was 60%- 70%. After filtration, the apheresis platelet met the leukocyte filtration standard,whereas manually-collected plated did not meet the leukocyte reduction standard. Filtration did not have significant impact on the function of platelet preparations stored in liquid, but the function of manual platelets was significantly lower than that of apheresis platelet. Conclusion Frozen stored platelet preparations are not suitable for leukocyte filtration due to the significant platelet loss.Currently the quality of manually concentrated platelets are inferior to that of apheresis platelet,and there is a need to develop quality leukocyte filters. The apheresis platelet is of good quality and could meet the leukocyte reduction standard using home-made filters, thus making it an ideal platelet preparation for leukocyte filtration.
