摘 要:目的探讨注射用血塞通对局灶性脑缺血再灌注神经生长因子(NGF)及酪氨酸激酶A(TrkA)含量变化的影响,并观察其对缺血性脑损伤的神经元保护作用。方法165只SD大鼠体重200~220g,随机分为三组:假手术组(n=55),局灶脑缺血再灌注组(n=55)和注射用血塞通治疗组(n=55)。线栓法制备大脑中动脉梗阻(MOAO)模型,缺血2h后恢复再灌注,药物组在脑缺血再灌注即刻和12h腹腔注射血塞通,模型组和假手术组均注射等量生理盐水。分别在缺血再灌注2、4、6、12h和24h处死动物,运用RT-PCR和免疫组化技术检测缺血侧顶叶大脑皮质NGF和TrkA的mRNA/蛋白的表达变化。结果(1)再灌注损伤后,脑皮质NGF和TrkA的mRNA表达均增高。在血塞通治疗组中NGF的mRNA于再灌注2h上升,6h达高峰;TrkAm RNA的表达在灌注2h也明显升高,但在4h达到高峰;(2)假手术组中NGF和TrkA的蛋白表达很低,再灌注损伤后,脑皮质TrkA的蛋白表达在再灌注12h和24h明显增高且血塞通治疗组高于脑缺血组和手术组,而NGF在再灌注24h明显增高且血塞通治疗组高于脑缺血组和手术组。结论脑缺血后神经元内NGF和TrkA的含量增多,可能有利于受损神经元的修复。注射用血塞通可能通过促进NGF和TrkA的表达而保护神经元。Objective To Investigate the effect Xuesaitong Injection on the expressions of nerve growth factor(NGF) and (Trk A) after focal cerebral ischemia/reperfusion(I/R) and study the neuroprotective effects of Xuesaitong Injection on ischemic neurons. Methods 165 Sprapgue Dawley rat weighing 200-220g were randomly divided into three groups: sham-operated group(n=55), I/R group (n=55) and Xuesaitong-treated group(n=55). Temporary middle cerebral artery occlusion (MCAO) model was applied. Immediately and 12h after reperfusion,rat received intraperitioneal injections of Xuesaitong Injection for the Xuesaitong Injection-treated group and the same volume physiological saline for the I/R group and sham- operated group. The expression of NGF and TrkA mRNA/protein were measured with methods of RT-PCR and immunohistochemistry respectively at 2, 4, 6, 12h and 24h after reperfusion in cerebral cortex of-rats. Results (1)The expression level of NGF and TrkA mRNA increased after reperfusion. In xuesaitong group, NGF mRNA increased significantly at 2 h after reperfusion, peaked at 6 h and TrkA mRNA increased significantly at 2 h after reperfusion, peaked at 4 h. (2) A very Low level of NGF and TrkA protein was detected in the cerebral cortex of sham group. The average gray values of TrkA positive product in xuesaitong group after 12 h and 24 h reperfusion were higher compared with sham group and IR group. The average gray values of NGF positive product in xuesaitong injection group after 12 h and 24 h reperfusion were higher compared with sham group and IR group. Conclusion NGF and TrkA maybe be related to the mechanism of neuronis injury and repairment. Xuesaitong injection may have protective effects on ischemic neurons by enhanced NGF and TrkA expression.
