摘 要:[目的]探讨山药多糖对小鼠肝、肾、心肌和脑组织体内外的抗氧化作用。[方法]昆明种小鼠分模型组、对照组、山药多糖高、中、低保护组,保护组每天按体重50mg/kg、100mg/kg、200mg/kg山药多糖灌胃,饲养8d,正常对照组腹腔注射调和油溶液,其余各组腹腔注射0.15%CCl4调和油溶液,24h分离血清测ALT、AST。用PBS制备肝、心、肾、脑匀浆;在组织匀浆中加入FeSO4和H2O2,诱导自由基的生成,测定各组织体内外的抗氧化作用。[结果]不同浓度的山药多糖保护组可降低血清中ALT和AST(P﹤0.05),降低各组织MDA的含量(P﹤0.01),体外抗氧化实验山药多糖能抑制脂质过氧化物的生成,A532nm明显降低(P﹤0.01)。[结论]山药多糖能降低CCL4损伤小鼠血清ALT、AST活性,对小鼠肝、肾、心肌、脑组织体内外有抗氧化作用,尤其对心肌、肾脏、肝脏抗氧化作用较强。[Objective] To investigate the anti-oxidant effect in vitro and in vivo of yam polysaccharide on mouse liver, kidney, and heart and brain tissues. [ Methods] Kunming mice were divided into model group, the control group, and polysaccharide protective group including high, medium and low-dosage of polysaccharide. Three polysaccharide subgroups were treated with gastric perfusion for 8 days by the body weight 50 mg / kg, 100mg / kg, and 200mg / kg of yam polysaccharide, respectively. At the end of the feeding, the control group was intra-peritoneal injected with blend oil solution, the remaining groups was intra-peritoneal injected with 0.15% CCl4 mixed with blend oil solution. After 24 hours, blood serum of mice was separated, and the ALT and AST were determined. The liver, heart, kidney and brain homogenate were prepared with PBS; FeSO4 and H2O2 were added to induce the generation of free radicals, and the anti-oxidant action of the tissues in vitro and in vivo were tested. [Results] Different concentrations of yam polysaccharides can reduce ALT and AST in serum (P﹤0.05) and lower the content of the MDA in tissue (P﹤0.01). Anti-oxidation experiments of yam polysaccharide in vitro can inhibit the production of lipid peroxidates and significantly decrease A532nm (P﹤0.01). [Conclusion] Dioscreapposite Thunb polysaccharide can reduce the activity of ALT and AST in serum on mouse’s liver damaged by CCl4. Besides, it is indicated that the polysaccharide show antioxidant on kidney, cardiac muscle, and brain tissue in vitro and in vivo, especially on the cardiac muscle, kidney and liver.
