摘 要:目的研究葛根黄酮对辛基酚染毒雄性大鼠生殖系统的影响。方法将60只健康清洁级雄性SD大鼠按体重随机分为对照组(灌胃同体积的玉米油)、辛基酚低剂量染毒组(80mg/kg)、辛基酚中剂量染毒组(160mg/kg)、辛基酚高剂量染毒组(320mg/kg),低剂量干预组(0.5g/kg葛根黄酮+320mg/kg辛基酚),高剂量干预组(5g/kg葛根黄酮+320mg/kg辛基酚),每组10只。采用灌胃方式进行染毒,每周3次,先给大鼠灌入辛基酚溶液,2h后,再给干预组大鼠灌入等体积的葛根黄酮溶液,灌胃体积为5ml/kg,连续60d。染毒后,取出睾丸,进行形态学观察,并统计精子数、活动度和畸形率。结果与对照组比较,高、中、低剂量染毒组和高、低剂量干预组的精子数和活动度均较低,畸形率均较高,差异均有统计学意义(P<0.05)。与低剂量染毒组比较,高剂量染毒组的精子数和活动度均较低,差异有统计学意义(P<0.05);且随着辛基酚染毒剂量的升高,精子数和活动度呈下降趋势,畸形率呈上升趋势。与高剂量染毒组比较,高、低剂量干预组的精子活动度均较高,畸形率均较低,高剂量干预组的精子数较高,差异均有统计学意义(P<0.05)。结论葛根黄酮对辛基酚存在着一定的拮抗作用。Objective To understand the antagonism af puerarin flavonoids (F) to octylphenol (OF) m the sperm damage. Methods Sixty male SD rats were randomly divided into three experimental groups (treated with octylphenol at doses of 80, 160,320 mg/kg) and two intervention groups (0.5 g/kg P+320 mg/kg OP, 5 g/kg P+320 mg/kg OP) and one control group. The administration was conducted by garage, 2 h after treated with octylphenol followed by puerarin flavonoids,three times a week for 60 consecutive days. Testicular morphological examination and sperm mobility were conducted. Results Compared with the control group,sperm count and mobility in the exposed groups and intervention groups were lower,malformation rate was higher(P〈0.05). Compared with the low-dose exposure group,sperm count and mobility in the high-dose exposed group were lower (P〈0.05). With increased dose of oetylphenol,sperm count and mobility presented a decreasing trend,malformation rate presented an increasing trend. Compared with the high-dose exposed group,sperm mobility in two intervention group was higher and malformation rates were lower. Sperm count in the high-dose intervention group was more than that in the high-dose exposed group (P〈0.05). Conclusion Puerarin flavonoids has an antagonism to octylpbenol in sperm damage.
