用两种PCR方法对HIV-1测定的敏感度对比分析

摘  要:目的:探讨用Roche公司生产的Combas Amplicor HIV-1病毒载量检测试剂与巢式RT-PCR法检测HIV-1这两种检测方法之间的相关性。方法:将81份HIV-1阳性患者血浆样本首先用Combas Amplicor病毒载量检测试剂盒检测出病毒拷贝数/ml,根据病毒拷贝数对数/ml的多少分为5组。再用巢式RT-PCR法对以上样本进行重复测定,观察以上2种检测方法的相关性。结果:用巢式RT—PCR法对用Combas Amplicor病毒载量检测的病毒拷贝数/ml分为2.000~2.999组;3.000~3.999组;4.000~4.999组;5.000—5.999组和〉6.000组病毒拷贝数/ml的检测阳性率分别为87.5%(7/8),93.75%(30/32),95.83%(23/24),100%(14/14)和100%(3/3)。结果表明,随着病毒拷贝数/ml的增高巢式法检测的阳性率也相应增高。结论:巢式RT—PCR由于具有操作简单,价格便宜等优点可作为AIDS的初步筛查,指导抗AIDS药物治疗等使用,对于基层而言是一种简便易行的方法。Objective : To investigate the relativity between quantitative detection of HIV-1 by Combas Amplicor virus load kit produced by Roche Company and qualitative detection of HIV-1 through nested RT-PCR method. Methods: 81 HIV-1 blood samples were obtained from HIV areas in Henan, first tested viral load copies/ml with quantitative Combas Amplicor, divided into 5 groups according to viral copies/ml then, re-tested above data by nested RT-PCR, observe the coincidence rate between this 2 Methods. Results: Compas Amplicor viral copies/ml as follows, 2. 000-2. 999 group; 3. 000-3. 999 group; 4. 000-4. 999 group; 5. 000-5. 999 group and above 6. 000 group, while each of the number of positive rate were 87.5% (7/8) ,93.75% (30/32) , 95.83% (23/24),100% (14/14) and 100% (3/3). The results indicate, with the viral copies/ ml become higher, the positive rate will be raise by nested RT-PCR. Conclusion: The teginning of period tested AIDS by nested RT-PCR, can guide therapy of at-AIDS, furthermore, it has advavtage of process simple, price inexpensive ,for the basic unit it is a method which can carry out.
