摘 要:目的探讨人脐带间充质干细胞(MSCs)移植到大鼠心肌后能否存活并向心肌样细胞分化,为心肌细胞的移植探索新的细胞来源。方法从人脐带华尔通氏胶分离、培养MSCs,检测人脐带来源的MSCs的细胞表面标记。用5-溴-2-脱氧尿苷(BrdU)标记人脐带间充质干细胞,标记细胞移植到梗塞大鼠心肌,观察移植后2周细胞能否存活,移植细胞能否向心肌样细胞分化,表达心肌细胞标记物troponinI及troponinT。结果人脐带间充质干细胞移植到心肌梗塞模型大鼠心肌后细胞能存活,并表达心肌细胞标记物troponinI及troponinT。结论人脐带间充质干细胞移植至体内能存活并分化为心肌样细胞,人脐带间充质干细胞可能作为心肌细胞移植的来源。Objective To differentiate human umbilical cord Wharton's Jelly-derived mesenchymal stem cells(MSCs) into cardiomyocytes in vivo, and to explore a new source of cells for transplantation of cardiomyocytes. Methods The MSCs labeled with BrdU were transplanted into cardiac muscle of the myocardial infarction rat model. The cell viability and the expression of troponinI and troponinT were observed 2 weeks later after the transplatation. Results The MSCs transplanted could still live 2 weeks later and express cardiac troponinI and cardiac troponinT. Conclusions The MSCs derived from human umbilical cord Wharton's Jelly can differentiate into cardiomyocytes in vivo. Based on these observations, and taking into account the immense advantages of UCB over other sources of stem ceils, the human umbilical cord-derived MSCs could retain myocardiogenic potential for treatment of intractable heart diseases.
