摘  要:目的探讨MMP-2、MMP-9基因mRNA在豚鼠实验性自身免疫性肌炎(EAM)肌肉组织中的表达和意义。方法实验分为正常组和EAM组。利用兔骨骼肌肌球蛋白加完全弗氏佐剂多次免疫豚鼠,同时联合腹腔注射百日咳毒素,制成EAM模型,观察其临床表现、血CK、肌肉病理的改变;用RT-PCR方法检测10例EAM豚鼠肌肉中MMP-2和MMP-9基因mRNA的表达,以10例正常豚鼠的肌肉组织作为对照组。结果 (1)MMP-2、MMP-9基因mRNA在EAM组肌肉中的表达较正常组显著增高(P<0.01)。(2)EAM组血CK的含量与MMP-2、MMP-9基因mRNA在肌肉中的表达程度呈正相关(P<0.01)。结论 MMP-2和MMP-9可能参与了EAM的发病。Objective To investigate the significance of the expression of gene mRNA of matrix metalloproteinases (MMP-2 and MMP-9 ) in the muscle of the exprimental autoimmune myositis (EAM) guinea pigs. Methods The EAM models was produced by repeatedly immunizating guinea pigs with rabbit skeletal muscle myosin plus Freunds complete adjuvant, and giving a celiac injection of pertussis toxin. The changes in clinical symptoms, serum creatine kinase and pathological profile were observed. The expression levels of MMP-2 and MMP-9 in the skeletal muscle were detected by RT - PCR,and were compared between EAM group(n = 10) and normal group ( n = 10). Results ( 1 ) There was significant upregulation of the expression of the MMP-2 and MMP-9 gene mRNA in the guinea pig muscle tissues of EAM group compared with normal group. (2) The content of serum creatine kinase was positively related to the level of the expression of the MMP-2 and MMP-9 gene mRNA in the muscle tissues of EAM group. Conclusions MMP-2 and MMP-9 may contribute to the onset of EAM.
