摘  要:将扩繁5代后的大花蕙兰类原球茎置于MS培养基中,以不同浓度水平的IBA,6-BA,活性炭和在缺少6-BA的情况下进行正交试验,观察类原球茎的平均增殖倍数和分化率,以及用高效液相色谱仪测定类原球茎中的6-BA,ABA,LAA,IBA,ZT,KT的含量。结果表明:适合于类原球茎增殖的培养基配方为MS+IBA 2.0mg/L+6-BA0.1~3.0mg/L+活性炭0.5-5.0g/L,适合于类原球茎分化的培养基配方为MS+IBA 0.1mg/L+6-BA 1.0mg/L4-活性炭2.0g/L。在多次继代培养的情况下,类原球茎的增殖和分化受到外源激素和内源激素的共同影响,已测到的高含量内源激素ZT,KT部分代替了外源激素6-BA起影响作用。The protocorm like-bodies (PLB) of Cymbidium hybridum after multiplying 5 generations were cultured in the MS medium containing different concentrations of IBA, 6-BA and active carbon, as well as the medium lack of 6-BA. The average multiplying times and differential rates of PLB were measured by the orthogonal test. The contents of 6-BA, ABA, LAA, IBA, ZT and KT in PLB were determined by HPLC. The results showed that the suitable medium formula for multiplying PLB was MS + IBA 2.0 mg/L + 6-BA 0.1 - 3.0 mg/L + active carbon 0.5 - 5.0 g/L and the suitable medium formula for differentiating PLB was MS + IBA 0.1 mg/L + 6-BA 1.0 mg/L + active carbon 2.0 g/L. In the process of multiple subculture, the multiphcation and differentiation of PLB were affected by exogenous hormones and endogenous hormones tngether. For example, the lack of 6-BA in the medium could be made up by the high content of endogenous hormones such as ZT and KT during multiplication and differentiation.
