黄芪、水蛭、水蛭素及水蛭黄芪配方含药血清对大鼠肾小球系膜细胞生长周期及凋亡的影响

摘　　要：目的:比较黄芪、水蛭、水蛭素及水蛭黄芪配方对大鼠肾小球系膜细胞(GMCs)生长周期、凋亡的影响及凋亡的形态学观察。方法:应用流式细胞仪检测GMCs生长周期与凋亡;用瑞氏染色,显微镜观察凋亡细胞的形态。结果:水蛭、水蛭素、黄芪、水蛭黄芪配方含药血清均能使大量GMCs处于G0/G1期,与对照组比较有显著性差异(P<0.01);均能提高GMCs凋亡率,水蛭、水蛭黄芪配方与对照组比较有显著性差异(P<0.01)。结论:水蛭、水蛭素、黄芪、水蛭黄芪配方含药血清均能阻止GMCs进入S期从而达到抑制增生的目的,且能提高GMCs的凋亡率,水蛭黄芪配方效果最优,水蛭次之。Objective： To investigate effect of the serum contained Radix Astragali, Hirudo, Hirudin and the compounding of these two herbs respectively on the growth cycle and apoptosis of rats＇ glomerular mesangial cells （ GMCs ） and observe the morphology of apoptosis. Methods： The growth cycle and apoptosis of GMCs was measured by flow cyt_ometer. Meanwhile morphous of apoptosis was visualized by Wright＇s staining and observed by microscope. Results： The serum contained Radix Astragali, Hirudo, Hirudin and the compounding respectively can keep most GMCs in the stage of G0/G1,and it had a significant deviation compared with the control（P〈0.01）.It also increased the rate of GMCs apoptosis, and it had a significant deviation compared with the control,too （P〈0.01）. Conclusion： The serum contained Radix Astragali, Hirudo, Hirudin and the compounding respectively can prevent GMCs entering the stage of S to inhibit its growth and it can also increase the rate of GMCs apoptosis.
