摘  要:目的研究妊娠中晚期SD大鼠的葡萄糖耐受能力及胰岛素分泌状况。方法选择妊娠15 d的SD大鼠作为实验组(n=6),以同批次未妊娠雌性SD大鼠作为对照组(n=6)。两组大鼠分别行腹腔内注射葡萄糖耐量试验(IPGTT)。原位胶原酶灌注法分离SD大鼠胰岛,倒置相差显微镜观察胰岛形态;放射免疫学法测定葡萄糖刺激后胰岛细胞的胰岛素分泌水平。结果IPGTT结果显示,实验组大鼠葡萄糖负荷后30、60、90和120 min时点的血糖值均明显低于对照组(P<0.05或P<0.01)。与对照组比较,实验组大鼠胰岛体积明显增大,胰岛细胞致密度增加。含16.7 mmol/L葡萄糖的KRB缓冲液刺激1 h时,实验组大鼠胰岛素分泌水平明显高于对照组(P<0.01)。结论妊娠中晚期SD大鼠的葡萄糖耐受能力及经葡萄糖刺激的胰岛素分泌能力均明显增强。Objective To investigate the glucose tolerance and ability of insulin secretion in SD rats in second and third trimesters of pregnancy. Methods SD rats with pregnancy of 15 d were selected as experimental group ( n = 6) , and another 6 rats of the same batch without pregnancy were served as controls ( n = 6). Intraperitoneal glucose tolerance trials (IPGTT) were conducted in these two groups. Rat islets were isolated after in situ collagenase digestion through pancreatic duct perfusion, islet morphology was observed by inverted phase contrast microscope, and insulin secretion was determined by radioimmunoassay. Results It was revealed by IPGTT that the levels of glucose at 30, 60, 90 and 120 rain were significantly lower in experimental group than those in control group( P 〈 0.05 or P 〈 0.01 ). Compared with control group, the islet volume and islet cell density are significantly increased in experimental group. The levels of insulin secretion in rats of experimental group after exposure to KRB buffer containing 16.7 mmol/L glucose for 1 h were significantly higher than those in control group (P 〈 0. 01 ). Conclusion Glucose tolerance and glucose-stimulated insulin secretion are significantly increased in SD rat islets in second and third trimesters of pregnancy.
