摘  要:以野大麦(Hordeumbrevisubulatum(Trin.)Link)的成熟胚为外植体诱导愈伤组织,选取淡黄色、易碎的胚性愈伤组织为材料,进行细胞悬浮培养.当建立起均质、分散且生长迅速的细胞系后,结合诱变剂甲基磺酸乙酯(EMS)对细胞的化学处理,在含盐0.8%NaCl和1.2%NaCl的MS培养基上筛选耐盐变异体.研究结果表明:筛选得到的变异体能在含盐培养基上持续增殖,而对照组则逐渐褐化死亡.变异体与对照组在游离脯氨酸含量、相对电导率、酯酶同工酶等几个生理生化指标方面存在着较大的差异.耐盐1.2%NaCl的愈伤组织的游离脯氨酸含量是对照组的6.68倍,而相对电导率比对照组小29.9%,两者的酯酶同工酶酶谱也存在明显的差异.以上分析表明,筛选出的愈伤组织具有一定的耐盐性且较稳定.耐盐愈伤组织转入分化培养基中,可经体细胞胚胎发生和器官分化两种途径再生植株,但分化率相对较低.Mature embryos of wild barley Horedum brevi subulatum (Trin.)Link were used as explants to induce callus.Yellowish and friable embryogenic calli were adopted to establish suspension cell cultures.After suspension cultures treated by ethylmethane sulfonate (EMS), sodium chloride tolerant variants were selected from them which cultured on saline medium.The results obtained from these studies showed that the variants could grow for a long time on 1.2% NaCl containing medium,while the controls died gradually.Free L-proline content of 1.2% NaCl tolerant variants was as much as 6.68 times of that of the control,and relative electrical conductivity ratio was 29.9% less than that of the control.Their esterose isozyme patterns were also different from the control with two additional bands,while their peroxidase isozyme bands were similar.These analyses showed that NaCl-selected calli had the characters of NaCl-tolerance.
