摘 要:目的:观察人参皂苷Rg1(Rg1)对缺糖缺氧所致C6胶质细胞损伤的保护作用及其机制。方法:采用MTT法检测了Rg1对缺糖缺氧所致胶质细胞C6的存活;采用流式细胞术检测了Rg1对胶质C6细胞凋亡的影响;采用W estern blot方法检测了Rg1对凋亡相关蛋白p38 MAPK磷酸化激活、P53、Caspase 3表达的影响,以及BDNF表达的影响。结果:Rg1通过抑制了缺糖缺氧所致C6细胞的凋亡而增加细胞的存活率,且抑制了p38 MAPK的磷酸化激活,抑制凋亡促进蛋白P53、Caspase 3的表达,并促进BDNF的表达。结论:Rg1可能通过抑制胶质细胞损伤而促进脑缺血中神经元的修复。Objective: To study the protective effect of ginsenoside Rg1 (Rg1) on the damage of C6 glioma cells induced by oxygen and glucose deprivation and its related mechanism. Methods: The cell viability was detected by MTT assay. The effect of Rgl on the C6 cell apoptosis was analyzed by flow cytometry. The phosphorylation of p38 MAPK, and the expression of P53, Caspase 3 and BDNF were detected by Western blotting. Results: Rg1 at 1 and 10 μmol.L^-1 remarkably increased the MTT reduction and inhibited C6 cell apoptosis after oxygen and glucose deprivation(P 〈0.05, P 〈 0.01). Rg1 inhibited the phosphorylation of p38 MAPK, and the expression of P53 and Caspase 3 ; it also promoted the expression of BDNF. Conclusion: Rg1 protects C6 cells from apoptosis induced by oxygen and glucose deprivation, which may contribute to its protective effect in cerebral ischemia.
