摘  要:To investigated physiochemical and biomechanical properties of our tissue-engineered decellularized porcine aortic valves and assessed their transmission risk of porcine endogenous retroviruses. Fresh porcine aortic valves were decellularized and its histological/biological characterization was determined. PCR and RT-PCR for detection of porcine endogenous retroviruses were performed in samples from fresh and decellularized porcine aortic valves and peripheral blood mononuclear cells from pigs and dogs bearing the porcine valve implants. In results, porcine aortic valves were completely decellularized with excellent preservation of extracellular scaffold structure. Shrinkage temperature and tensile strength were similar in decellularized porcine aortic valves and controls. Decellularized porcine aortic valves showed good capability to support repopulation of human endothelial cells. No porcine endogenous retrovirus was measurable in decellularized porcine aortic valves and dogs peripheral blood mononuclear cells. The decellularized porcine aortic valves offer physiologically functional, mechanically durable, and virus-free tissue-engineered heart valves for possible use in clinical applications.
