摘 要:目的观察大黄对脓毒症大鼠血清中高迁移率族蛋白B1(HMGB1)、肿瘤坏死因子-α(TNF—α)的影响,揭示大黄治疗脓毒症的机制。方法采用盲肠结扎穿孔术(CLP)制备脓毒症模型,104只雄性SD大鼠随机分为正常对照组(8只)、CLP组(48只)、大黄治疗组(48只)。分别在术后0、3、8、24、48及72h活杀大鼠留取血标本,采用酶联免疫吸附法测定TNF-α和HMGB-1的含量。结果大黄治疗组血浆TNF-α水平在8h,HMGB1水平在24、48及72h明显低于CLP组,差异有统计学意义。结论大黄通过降低血清中TNF—α和HMGB-1的水平,对脓毒症大鼠具有治疗作用。Objective To observe the influence of rhubarb preparation on high mobility group box 1 protein ( HMGB1 ) and tumor necrosis factor - α( TNF - α ) in rats with sepsis, and investigate the mechanism of rhubarb on sepsis. Methods A sepsis model reproduced by cecal ligation and puncture (CLP) ,and 104 male SD rats were randomly divided into normal control group( n = 8) ,CLP group ( n = 48 ) , the rhubarb treatment group ( n = 48 ). The rats were killed at 0,3,8,24,48 and 72 hours after operation and the Plasma was sampled. The contents of TNF - α and HMGBI were detected by enzyme linked immuneoadsorbent assay. Result TNF - α level of plasma were significant increased at 8h post - CLP compared with rhubarb treatment group. In rhubarb treatment group, HMGB - 1 level decreased in different degrees compared with CLP group at 24,48,72 h. Conclusions Rhubarb possesses the action of treat- ment in rats with sepsis by decreasing the TNF-α and HMGB1 contents.
