医药中间体3,4-二羟基-5-甲氧基苯甲酸甲酯的合成工艺改进

摘　　要：3,4-二羟基-5-甲氧基苯甲酸甲酯是一种肝脏类医药的中间体。本文采用新工艺合成了3,4-二羟基-5-甲氧基苯甲酸甲酯,优化了工艺条件。实验结果表明:3,4-二羟基-5-甲氧基苯甲酸甲酯的最佳条件为:n(甲醇):n(没食子酸)=3.0:1,n(Me2SO4):n(没食子酸)=1.15:1,浓硫酸用量为1.7mL,氢氧化钠用量为12g,硼砂用量为36g。在此条件下,总收率可达78.3%,提高了14.4%。因此,合成3,4-二羟基-5-甲氧基苯甲酸甲酯具有重要的理论意义和现实价值。3,4 -Dihydroxy -5 -methoxy methyl benzoate was a very important liver medical intermediate. The news synthetic process of 3,4 -dihydroxy -5 -methoxy methyl benzoate was introduced on this paper. And the synthetical technology was optimized. The results showed the best conditions for synthesis of 3,4-dihydroxy-5-methoxy methyl benzoate were that：n （methanol）：n（gallic acid）=3.0：1,n （Me2SO4）：n （gallic acid）=0.7：1,the dosage of vitriol, sodium hydroxide and borax were 1.7mL, 12g and 36g respectively.Under these conditions,the total yield of the products which was 14.4% higher than existing technology reached 78.3%. So, its synthesis had very important theoretic significance and practical value .
