摘 要:目的研究重组人白细胞介素-1β(rhIL-1β)对正常人外周血单核细胞(PBMC)吞噬功能及产生肿瘤坏死因子-α(TNF-α)和前列腺E2(PGE2)的影响及芍药苷(Pae)的作用,并探讨部分机制。方法Ficoll密度梯度离心法及贴壁法分离PBMC,依次加入不同浓度rhIL-1β培养24h或同一浓度rhIL-1β培养不同时间及不同浓度Pae作用24h,收集培养细胞及上清液,中性红染色法观察PBMC的吞噬功能,放射免疫法(RIA)和酶联免疫吸附法(ELISA)分别测定TNF-α和PGE2的水平。结果体外给予rhIL-1β(0.01、0.1、1、10μg/L)可不同程度提高正常人PBMC的吞噬功能及TNF-α和PGE2的产生水平;体外给予1μg/LrhIL-1β(3、6、12、24h)可使正常人PBMC产生TNF-α和PGE2的水平升高;Pae(10-9、10-8、10-7、10-6、10-5mol/L)体外作用可不同程度抑制rhIL-1β(1μg/L)对正常人PBMC的促吞噬作用及TNF-α和PGE2的产生。结论rhIL-1β体外作用可增强正常人PBMC的吞噬功能,升高TNF-α和PGE2水平;Pae体外作用可抑制PBMC增强的吞噬功能,降低TNF-α和PGE2的产生水平。Objective To investigate the effects of recombinant human interleukin-1β (rhIL-1β ) on phagocytosis function and tumor necrosis factor-α(TNF-α) and prostaglandin E2 (PGE2 ) production of peripheral blood mononuclear cells (PBMC) from healthy volunteers and the role of paeoniflorin (Pae). Methods PBMC was gained by density gradient centrifugation and incubated with different concentration of rhIL-1β for 24 hours or rhIL-1 β ( 1μg/ L) for different time, and different concentration of Pae was added to culture 24 hours. Then cultured cells and supernatant fluids were collected. Phagoeytosis function was measured by neutral red staining, and the levels of TNF-α and PGE2 were detected by radioimmunoassay (RIA) and enzyme linked immunosorbent assay (ELISA) respectively. Results rhIL-1β(0. 01, 0. 1, 1,10 μg/L) could promote phagocytosis function and increase TNF-α and PGE2 production of PBMC; rhIL-1β 1 μg/L (3, 6, 12, 24 h) increased the levels of TNF-α and PGE2. Pae ( 10 ^-9,10 ^-8,10 ^-7,10 ^-6,10 ^-5 mol/L) in vitro suppressed the stimulation of 1 μg/L rhIL-1β on PBMC, manifesting the down-regulation of phagocytosis function and TNF-α and PGE2 production. Conclusion rhlL-1β could promote phagocytosis function and increase the levels of TNF-α and PGE2 of PBMC. Pae shows a capacity to reverse the effects of rhIL-1β on phagocytosis and TNF-α function and PGE2 production of PBMC.
