摘 要:将采集的山羊精液去除精清,分为鲜精组、冻精组、反复冻融致死组3组进行处理后,与地高辛标记的线性化PCS2EGFP质粒共育转染,用免疫组化的方法检测精子转染外源DNA的效率.结果显示鲜精、冻精和反复冻融致死3种处理组的阳性转染效率分别为(36.61±7.91)%,(32.68±3.38)%,(69.36±4.79)%.反复冻融致死组转染率极显著高于鲜精组和冻精组(p<0.01),鲜精组和冻精组转染效率差异不显著(p>0.05).Goat semen was centrifuged and divided into three groups:fresh sperm, frozen-thawed sperm and dead sperm by repeated frozen-thawed treatment. The sperms were incubated in vitro with foreign DIG-labeled linear PCS2EGFP plasmid. The transfection rates of the sperms were determined immunohistochemically. The results showed that the sperm DNA-binding rates were (36.61±7.91)%, (32.68±3.38)% and (69.36±4.79)%, respectively, for the above-mentioned three groups, and the sperm DNA-binding rate of dead sperms with repeated frozen-thawed treatment was significantly higher than that of the other two groups (p〈0.01).
