摘 要:目的观察大鼠去胆管及去门静脉肝叶蛋白质组变化,探讨去胆管肝叶的保留价值。方法实验大鼠随机分成5组:BL组,单纯栓塞并结扎右叶胆管观察1个月;PL组,单纯结扎方叶门静脉观察1个月;BPL组,栓塞结扎肝右叶胆管,同时结扎方叶门静脉,并分为2周、1个月和2个月观察组。提取大鼠去胆管及去门静脉肝叶肝组织总蛋白质并进行双向电泳分离及质谱鉴定分析,比较自身对照两肝叶白蛋白、载脂蛋白A1、磷酸丙糖异构酶、Z蛋白和Y蛋白5种代谢相关性蛋白质变化的差异。结果双向电泳凝胶图像分析每块胶平均识别蛋白点1 496±150个,有显著性差异的蛋白质点达到87.2±31.2个。自身两个对照肝叶平均匹配率达57.6%左右。去胆管肝叶白蛋白、载脂蛋白A1、磷酸丙糖异构酶表达无明显降低,生物转化功能蛋白Z蛋白和Y蛋白明显降低;去门脉肝叶表现正好相反。结论在去胆管和去门脉条件下,肝细胞存在明显的功能分化现象;去胆管肝叶保留了大部分蛋白合成分泌及营养物质代谢功能,更具有保留价值。本实验为临床特殊情况的处理(如某些良性胆道疾病仅处理胆管而保留相应肝叶肝脏组织)提供了初步的实验依据。Objective To explore the preservation value of bile duct deprived (BDD) hepatic lobe by observing changes in proteome of rat liver tissue. Methods Rats were divided into five groups as follows: Group BL with the right lobe bile duct embolized and ligated for one month, Group PL with the quadrate lobe portal vein ligated for one month, and three BPL Groups with the fight lobe bile duct embolized and ligated as well as the quadrate lobe portal vein ligated. The observation time points were set as two weeks, one month and two months. The total proteins of BDD lobes and portal vein deprived (PVD) lobes of liver tissue were extracted and then separated and identified by two-dimensional gel electrophoresis (2-DE) and mass spectrometry. Five metabolism relevant functional proteins (i. e. albumin, apolipoprotein A-1, triosephosphate isomerase, Z protein and Y protein) were specifically compared with that of the self-control hepatic lobes. Results Analysis of 2-DE gel showed: 1 498-7150 protein spots were seen in the two self-control gels and 87. 2±31.2 differential protein spots were revealed. The average match rate was about 57. 6%. Proteomics study revealed that the expression of albumin, apolipoprotein A-1 and triosephosphate isomerase in the BDD lobe showed no obvious reduction, while the Z and Y proteins decreased significantly. The PVD lobe showed a reverse change. Conclusions Under the condition of BDD and PVD, liver cells may show a significant functional differentiation. The BDD lobe retains most of the liver functions, such as protein synthesis and nutrition metabolism, and it is the refore of preservation value. The present study provides a preliminary experimental basis of preserving the BDD liver lobe in some emergency and specific clinical situations.
