摘　　要：目的探讨IL-6对P815肥大细胞蛋白酶激活受体(PAR)表达的影响及其在过敏与炎症中的作用机制。方法P815肥大细胞激发后,收集各时间点的细胞和上清。采用实时定量PCR和流式细胞术的方法在mRNA和蛋白水平检测PAR-1、PAR-2、PAR-3和PAR-4表达情况。结果IL-6上调PAR-1、PAR-2、PAR-3、PAR-4mRNA的表达;下调PAR-1、PAR-3、PAR-4蛋白的表达;IL-6抗体的应用可阻断IL-6对PARs表达的上调作用。结论IL-6能够调节肥大细胞表面PAR的表达,并可能通过这一途径参与过敏性炎症反应。Objective To investigate the Modulation of mast cell proteainse activated receptor expression by IL-6 and it＇ s mechanism of inflammation. Methods After being challenged, some of P815 cells were treated and analyzed by flowcytometry to detect expression of PRAs at protein level, some of cells were treated and analyzed by real time RT-PCR to detect expression of PARs at mRNA level. Results The results showed that IL-6 upregulated mast cell PAR-1, PAR-2, PAR- 3, PAR-4 mRNA expression and downregulated PAR-1, PAR-3 ,PAR-4 expression at protein level, meanwhile,this modula- tory action of IL-6 were blocked by administration of IL-6 blocking antibody. Conclusion IL-6 can regulate mast cell PARs expression and may contribute to the process of inflammation by this way.
