摘  要:[目的]探讨白皮松落针病病原菌的生物学特性。[方法]研究了不同培养基、pH值、温度对其病原菌——白皮松散斑壳菌落生长的影响,并对其ITS序列进行了分析。[结果]病原菌菌丝在4种培养基上均能生长,并以OA上生长最快;菌丝生长适宜pH值为5.5~6.5,最适pH值为6;生长适温为22~26℃,最适温度为25℃。利用通用引物ITS4和ITS5进行PCR扩增,得到的rDNA ITS序列长度为443 bp,其中包括125 bp的ITS1序列、169 bp的5.8S rDNA序列和149 bp的ITS2序列。[结论]为白皮松落针病的准确诊断及分子鉴定提供理论依据。[Objective] The aim was to study biological characteristics of the needle cast pathogen of lacebark pine.[Method] The effects of medium,pH and temperature on colony growth of Lophodermium pini-bungeanae were studied,and internal transcribed spacer(ITS) sequence of Lophodermium pini-bungeanae was analyzed. [Result] The results showed that the mycelium could grow on four kinds of culture media including MA,OA,PA and PDA,and OA was the best one.The suitable pH values ranged from 5.5 to 6.5,and the optimum pH was 6.The proper temperature ranged from 22 ℃ to 26 ℃,and the optimum temperature was 25 ℃.ITS4 and ITS5 primers were used to amplify the rDNA-ITS regions of the pathogen by the polymerase chain reaction(PCR),443 bp rDNA-ITS regions were obtained,which included ITS1 of 125 bp,5.8S rDNA of 169 bp and ITS2 of 149 bp.
