摘  要:利用化学恒温培养箱对小鼠实施42℃1 h/d的热应激处理,建立持续的全身热应激动物模型。在热应激持续到8、13、21和35 d时,颈椎脱臼处死小鼠,应用免疫荧光组织化学和蛋白质印迹分析的方法检测睾丸组织Hsp70的表达。结果显示,对照组的小鼠睾丸组织Hsp70呈极强表达;热应激组小鼠睾丸组织Hsp70表达减弱,但随着热应激持续时间的延长,Hsp70表达逐渐恢复。热应激处理8 d后的小鼠睾丸组织Hsp70呈弱表达、13和21 d后呈极弱表达、35 d后呈弱表达。结果表明,Hsp70在小鼠睾丸正常的精子发生过程中发挥重要的作用;持续性的热应激可以使机体建立热适应,Hsp70可作为睾丸组织对热应激产生热适应的生物学标志之一。Mice were treated by heat stress at 42 ℃ for one hour per day by using incubator to establish the continous systemic heat stress animal model.The mice in control group and heat stress group were killed by cervical dislocation method,when they were treated by heat stress for 8,13,21and 35 d.And the expression of Hsp70 in the testis tissue was detected by immunofluorescence histochemistry and western blot analysis methods.The results showed that there was a strong expression of Hsp70 in control group.The expression of Hsp70 in heat stress group was weakened,however,the expression of Hsp70 recovered gradually with the prolongation of heat stress time.The expression of Hsp70 with heat stress for 8 d was weak,and that was very weak after 13 and 21 d and that was weak after 35 d.The results indicated that Hsp70 played an important role in normal spermatogenesis process of mice testis.The sustained heat stress could establish heat adaptation in the body.And Hsp70 could be used as one of biological markers that testis produced heat adaptation to heat stress.
