摘  要:目的建立胎鼠神经干细胞体外培养方法并鉴定神经干细胞,观察神经干细胞生长特点。方法采用含碱性成纤维细胞生长因子和表皮生长因子的无血清培养基对大鼠胚胎间脑组织细胞悬液进行培养。通过检测神经干细胞的特异性抗原、神经干细胞的自我增殖能力和分化能力来鉴定神经干细胞。结果分离培养出了大量具有不断增殖的能力、表达神经巢蛋白的神经干细胞,并能经过诱导分化为神经元和神经胶质细胞,并能稳定传代20代以上。结论无血清培养基分离培养的胎鼠脑组织神经干细胞具有自我更新和增殖能力,巢蛋白细胞免疫荧光鉴定为神经干细胞,成功建立了大鼠神经干细胞的分离培养方法。Objective To establish the methods of isolation, culturing of neural stem cells (NSC) in vitro and observe the characteristics of NSC growth. Methods NSC were isolated and cultured by microdissection, mechanical blowing and serum-free suspension culture. The cells were identified by detecting the differential NSC antigennestin,the ability to proliferate continuously and differentiate into neurons and glial ceils by inducement. Results The cultivated cells have the ability to proliferate continuously. They expressed nestin protein and can differentiate into neurons and glial cells by inducement. They have reproduced for above 20 generations. Conclusion The method can be used to isolate and culture NSC from embryonic rat brain, the cell identified as NSC possess the ability of proliferation and self-reproduction. The method of isolation and culture of rat NSC were successfully established.
