中药方剂对不同证型荨麻疹患者PBMC中细胞因子IL-4、IFN-γ表达的影响

摘　　要：目的探讨消风散、玉屏风散、当归饮子对三型荨麻疹患者外周血单个核细胞(PBMC)中白介素-4(IL-4)、干扰素-γ(IFN-γ)表达的影响。方法采集68例荨麻疹患者及24例正常人静脉血,加入不同浓度10-1～10-7g/ml中药或RPMI-1640进行体外孵育,采用双抗体夹心ELISA方法检测IL-4、IFN-γ水平。结果荨麻疹各型患者IL-4细胞因子水平均高于正常人;IFN-γ细胞因子水平均低于正常人;各型荨麻疹患者PBMC经各代表方剂干预后IFN-γ的含量升高,且差异有统计学意义(P<0.05);IL-4的含量较空白组略改变,差异无统计学意义(P>0.05)。结论各中药方剂可能通过提高IFN-γ水平,使相应证型荨麻疹患者Th1/Th2失衡状态均得到一定程度的纠正,从而达到治疗作用。Objective To investigate the effects of Xiaofengsan,Yupingfensan and Dangguiyinzi decoction on the expression of interferon-γ （IFN-γ） and interleukin-4 （IL-4） of peripheral blood morphological cells （PBMC） in urticaria. Methods Venous whole blood was obtain from 68 patients with urticaria and 24 healthy human subjects,and cultured in vitro with three kinds of decoctions at different density from 101 g / ml to 107 g / ml or with RPMI-1640 respectively. Double antibody sandwich ELISA was used to detect the levels of IFN-γ and IL-4. Results The levels of IL-4 in all patients with urticaria were higher than those in normal person,and the levels of IFN-γ were lower. The levels of IFN-γ increased significantly after adding three kinds of decoction（P〈0.05） ,but the levels of IL-4 have no significant difference,compared with the control group （P〈0.05）. Conclusion Chinese medicinal formulae can adjust the imbalance of Thl/Th2 by enhancing the secretion of IFN-γ to treat urticaria.
