从复方中药糖肾胶囊干预IR大鼠的研究浅谈实验动物模型及实验方法  被引量:2

摘  要:目的比较和探讨分别以高果糖餐、高脂肪餐诱导的IR大鼠模型的相关指标及实验方法。方法120只清洁级SD大鼠,随机分为高果糖餐组和高脂肪餐组,每组内又分为正常组(普通餐)、实验组,成模后,实验组分为实验对照组、糖肾胶囊组和文迪雅组,连续灌喂4周,比较各组体重、血压、血糖、血脂、M值。结果以高果糖餐造模的大鼠表现为消瘦、高血压的IR模型;以高脂肪餐造模的大鼠表现为肥胖、高血压、脂代谢异常的IR模型,接近代谢综合征的特点。复方糖肾胶囊对两组模型大鼠的IR、血压等指标均有一定改善作用。结论两组模型均为成功且稳定的IR模型,其中高脂肪餐大鼠模型亦可用于代谢综合征的研究;传统中药煎剂中的多种复合成分针对IR的多靶点的综合调节优于单一的中药提取物和西药的单一靶点的调节,并提示我们传统中药水煎剂中所含的主要成分之外的一些微量成分的重要作用。Objects Compare and discuss the indicators and experimental methods about insulin resistant models of fructose-fed-rats and fatty-diet-fed rats. Methods 120 SD rats were randomly divided into fructose-fed-rats and fatty-diet-fed rats groups. Each group were than divided into control group, experimental group, Tangshen-Capsule group and avandia group. Than compare the body weight, blood pressure, blood sugar, blood lipid and M feature. Results The model of fructose-fed-rats show the characters of thin, hypertension and IR. The model of fatty-diet-fed-rats show the characters of fat, hypertension, dyslipidemia and IR. The second model close to the metabolic syndromes. Tangshen-Capsule can improve IR and blood pressure of both models. Conclusion Both models are successful and stable IR models. The fatty-diet-fed-rats model can be used in research of metabolic syndromes. The integrated functions of traditional Chinese medicine is superior to the single target of western medicine. This remind us that some micro dose ingredients in traditional Chinese medicine may act great roles in treating.
